Metabolomics is a rapidly developing approach to the study of metabolite composition in biological systems. However, most metabolomic studies to date have been conducted on model systems, such as bacterial, fungal or yeast cultures, with relative levels of metabolites measured by NMR spectroscopy and other physicochemical methods. Whilst much attention has been given to metabolic profiling in biological fluids and cells, very few studies have been conducted on tissues. Here we present a study of the major human colonic metabolome by high resolution 1H NMR spectroscopy. The colonic metabolome was characterized by the presence of several high molecular weight molecules, including maltodextrins, lactulose, and starch. In addition to structural identification of metabolites by homonuclear 1H NMR spectroscopy, we were able to identify each of the polyglucoses using the anomeric region of the 1H NMR spectrum, characteristic of many types of sugar. This was made possible by introducing specific 1H pulse sequences to detect these resonances. Furthermore, the presence of small amounts of glucose was detected in the colon using 1H NMR spectroscopy, without the need for derivatization. In addition to the polyglucoses, lower molecular weight metabolites, such as acetate, lysine, valine, tryptophan, and glucose were detected, which we believe can be attributed to microbial metabolism.
